Sample preparation: chemical fixation (2.5 % glutaraldehyde, 4 % formaldehyde, 0.5% NaCl), rOTO embedding, Durcupan
FIB-SEM: SEM: 1.5 kV, ESB grid 450V, pixel size x/y 5 nm, continuous mill-and-acquire mode; FIB: 3nA), z-step 50 nm
Postprocessing: alignment with Fiji/ImageJ TrackEM2, cropped, inverted and blurred (Gaussian blur, sigma 2) with Fiji/ImageJ
