Unaligned multi-frame micrographs of Bos taurus mitochondrial complex I from an as-prepared sample solubilised in DDM and reconstituted in to MSP2N2 nanodiscs. Frozen on 0.6/1 UltrAuFoil grids (though holes were larger). The grid was treated with a PEG-thiol reagent (11-mercaptoundecyl hexaethyleneglycol) beforehand to increase hydrophilicity. Images (TIFF) were collected with a 300 keV FEI Titan Krios on a Falcon K3 detector, using counting mode with super-resolution mode and a Gatan energy filter (20 eV). The C2 and objective lens apertures were set to 70 and 100 µm, respectively. A defocus range of -1.0 to -2.4 was used. 2.4 s exposures were captured at a dose rate of 16.9 electrons Å−2 s−1 in 40 frames with a total dose of 40.5 electrons Å−2. A pixel size of 0.535 Å/pixel (super resolution; nominal pixel size was 1.07 Å/pixel) was used. Datasets with and without AFIS were collected. XML metadata are also included, as are gain reference files and pixel defect files.
