Real-time capture of σN transcription initiation intermediates reveals mechanism of ATPase-driven activation by limited unfolding
Raw micrographs were collected on a Talos Arctica at 200kV equipped with a Gatan K2 camera. Recordings were performed in counting mode at a pixel size of 1.5 A/px with a dose rate of 8.4 electrons/px/s for an exposure time of 12 seconds in dose-fraction mode of 0.3 seconds per-frame resulting in a total of 40 frames and 1.12 electrons/A^2/frame (total dose of 44.8 electrons/A^2). Defocus range for the collection was -0.8 to -3.0 microns. The tilt angle of the specimen was 0 degrees.
Raw micrographs were collected on a Talos Arctica at 200kV equipped with a Gatan K2 camera. Recordings were performed in counting mode at a pixel size of 1.5 A/px with a dose rate of 9.2 electrons/px/s for an exposure time of 12 seconds in dose-fraction mode of 0.3 seconds per-frame resulting in a total of 40 frames and 1.227 electrons/A^2/frame (total dose of 49.1 electrons/A^2). Defocus range for the collection was -1 to -2.5 microns. The tilt angle of the specimen was 20 degrees.
0 degree tilt data. Raw movies of were aligned using MotionCor2 (through Relion) with a binning factor of 1 and a dose of 1.12 electrons/A^2/frame. Full job output is provided in the upstream directory.
