Volta phase plate with defocus cryo-EM dataset of Thermoplasma acidophilum 20S proteasome [427 multi-frame micrographs composed of 24 frames each in TIFF format]
Publication:
Using the Volta phase plate with defocus for cryo-EM single particle analysis
The movies were acquired with SerialEM as dark-subtracted but non-gain-normalized stacks and saved as LZW-compressed TIFFs. The gain reference is in the "SuperRef_20S_000_Mar28_14.49.23.mrc" file. "MotionCor2_patch9_DW1.sh" is an example shell script for using MotionCor2 to gain normalize, align and dose-weight the movies. "micrographs_ctf.star" is a Relion 2.0 .star file containing the CTF fit results from ctffind4 v4.1.5. "particles.star" is a Relion 2.0 .star file containing particle coordinates and CTF fit parameters. Other details about the dataset are in "dataset_parameters.txt". The data was used for three reconstruction strategies. All frames were used in the Relion particle polishing and Warp approaches. The first two frames were discarded in the MotionCor2 approach.
