Optic nerve of a conditional and inducible Mbp knock-out mouse (Mbpfl/fl*PlpCreERT2+),16 weeks after tamoxifen-induction at the age of 8 weeks
Sample preparation: HPF and freeze substitution, embedding in EPON
FIB-SEM: SEM: 1.5 kV, ESB grid 450V, pixel size x/y 5 nm, continuous mill-and-acquire mode; FIB: 3 nA, z-step 50 nm
Postprocessing: alignment with Fiji/ImageJ TrackEM2, cropped, inverted and blurred (Gaussian blur, sigma 2) with Fiji/ImageJ
